INTRODUCTION: THE VALUE OF AUTOMATED DATA ANALYSIS

With traditional flow cytometers, operators manually analyze the data by sequentially applying positive and negative thresholds or gates around cell events in a series of two-dimensional plots, drilling down to discriminate each cell subset population defined by a unique combination of fluorescently labeled markers and light scatter properties.

Analysts must be able to navigate and analyze a wide range of staining patterns and cell subset frequencies, especially in clinical samples from patients with different disease states and prior treatment regimens. Emphasizing the impact of such biological variability, a meta-analysis of stem cell therapy starting material shows that the final therapeutic dose could vary across patients by up to four orders of magnitude, creating significant challenges when trying to analyze results.3 This requires specialized skills and knowledge to correctly discriminate and classify subpopulations of cells.
